Sequence analysis of fruiting bodies isolated from Ash trees at Ashwellthorpe lower wood

The material

We used DNA from fruiting bodies growing on dead ash racines and amplified the ITS region of 18S rDNA using universal primers: 5’-TCCGTAGGTGAACCTGCGG-3’ and 5’-TCCTCCGCTTATTGATATGC-3’

The analysis

The DNA sequences were blasted against the NCBI nucleotide database.

The interpretation

The tblastn search identified two types of fruiting body

1) 92% identity (683/745) with Typhula phacorrhiza 18S rRNA

2) 97% identity (486/501) with Lachnum pudibundum 18s rRNA

This is the first report of these two fungal organisms at Ashwellthorpe lower wood.
